Abstract
The heart derives the majority of its energy requirements from the oxidation of fatty acids and glucose. During states of nutritional deficiency, cardiac reliance of the oxidation of fatty acids increases at the expense of glucose oxidation. Since a key rate-limiting step of fatty acid oxidation is carnitine palmitoylcarnitine type 1 (CPT-1) which is regulated by malonyl CoA levels, we examined whether the expression levels of the enzymes that regulate the levels malonyl CoA are increased during nutritional deficiency. We demonstrate that the increase in fatty acid oxidation noted during fasting is not noted in the presence of an increased expression of acetyl CoA carboxylase (ACC), malonyl CoA decarboxylase (MCD) or 5' AMP activated protein kinase (AMPK). Additionally, we examined whether 'in vivo' intracoronary delivery of constitutively active AMPK would result in increased fatty acid oxidation. However, due to the low efficiency of gene delivery noted following intracoronary delivery, we were unable to determine the metabolic effects of this over-expression and the study evolved to one where different gene delivery techniques were examined for efficiency of gene delivery to the myocyte.
