Abstract
In hybrid poplar ('Populus trichocarpa' * ' P. deltoides'), mechanical wounding, FTC herbivory, and methyl jasmonate are known to induce expression of polyphenol oxidase (PPO) and trypsin inhibitor (TI). To identify novel wound-responsive genes, a suppressive subtractive hybridization (SSH) library was constructed from young leaves showing strong systemic-induction of PPO and TI after wounding. Macro-arrays were differentially screened using 32P-radiolabelled cDNA representing total mRNA from mechanically wounded, FTC browsed, and control leaves. Wound-induced cDNAs were sequenced and their predicted peptide sequences were used to query databases to predict their function. Induction locally and systemically was confirmed in poplar leaves after mechanical wounding. Herbivory was compared to mechanical wounding and the chemical elicitors methyl jasmonate and salicylic acid. Unwounded poplar tissues were examined to explore developmental gene expression of the wound-induced genes. The results presented in this thesis have expanded the number of known wound-induced genes in the poplar system.
