Abstract
Xenobiotics are recalcitrant to microbial degradation, the usual treatment for industrial and municipal wastes. Biphasic fermentation, which involves partitioning a substrate dissolved in an immiscible solvent into the aqueous phase at sub-inhibitory concentrations, can circumvent the inherent problems of xenobiotic degradation--namely substrate inhibition and toxicity--resulting in improved degradation rates. This work involves the development and investigation of a biphasic fermentation system for the degradation of pentachlorophenol (PCP) by Arthrobacter sp. ATCC 33790. Using systematic selection criteria, diethyl sebacate was chosen as the biphasic solvent. The batch biphasic fermentation of PCP achieved near complete removal of 10 g of PCP with volumetric consumption rates of 82-84 mg/L-h, more than doubling previously published rates. However, gas chromatography and mass spectrometer analysis indicated the formation of a degradation intermediate. Although it could not be positively identified, the compound was not believed one of the known biodegradation pathway intermediates.
