Abstract
Four proteins (FAT/CD36, FABPpm, FATP1 and FATP4) are known to play a role in the transport of long chain fatty acids (LCFA) into mammalian skeletal muscle. Growing evidence suggests that FAT/CD36 and FABPpm function cooperatively. In the current study, the relationship between these proteins was further examined by transiently over-expressing the FABPpm-FLAG construct in two skeletal muscle cell lines, one expressing CD36 (mouse-derived C2C12 cells), and one without detectable levels of the protein (rat-derived L6 cells). Despite low transfection efficiency, the transgene was expressed on the cell surface of both L6 and C2C12 myotubes, as shown by immunofluorescence. Palmitate uptake assays, used to evaluate the functional effects of transgene expression, indicated that over-expression of FABPpm significantly (P<0.05) increased the initial rate of LCFA transport in the CD36-positive C2C12 cells, whereas transport was unchanged in the L6 myotubes, suggesting that FABPpm only functions to transport LCFA when in the presence of CD36.
