Abstract
In a yeast two-hybrid screen, the Rvs167 protein was found to interact with the Pho85-associated G1 cyclin, Pcl2. RVS167 encodes an SH3 domain-containing protein implicated in cytoskeletal reorganization in response to starvation and plays a role in the bud site selection mechanism. Using genetic analysis and affinity chromatography, Rvs167 protein was shown to associate with both Pcl2 and Pho85, possibly via the formation of an Rvs167-Pcl2-Pho85 complex. Preliminary in vitro binding data suggests that Rvs167 may also associate with two other cyclins involved in cell cycle-related processes. These cyclins are Cln1 and Ccl1. These interactions do not require the SH3 domain of Rvs167, consistent with the presumed involvement of the SH3 domain in actin cytoskeleton association. Like rvs167 deletion mutants, pho85 deletion mutants display a reduction in viability upon starvation and a budding defect in diploids. These results suggest that Rvs167 and Pho85 may be partners in the coordination of nutritional signals with morphogenesis and cell proliferation.
